Sabotaging the breaks: FSEN1 expands the toolbox of FSP1 inhibitors
The identification of increased lipid peroxidation has highlighted vulnerabilities in challenging cancers. In this context, FSP1 has emerged as a crucial regulator, counteracting the damaging effects of lipid peroxidation. In a groundbreaking study published in Cell Chemical Biology, Hendricks et al. introduce FSEN1, a powerful inhibitor of FSP1. The discovery of FSEN1 offers great potential, as it enhances the effectiveness of GPX4 inhibitors and several FDA-approved drugs, boosting their ability to induce ferroptosis in resistant cancer cells. This marks a significant advancement in the development of ferroptosis-based strategies for preclinical cancer treatments.
